Overview

JP1201 is a small molecule Smac (Second Mitochondrial-derived Activator of Caspases) mimetic developed by Joyant Pharmaceuticals. It functions as an antagonist of Inhibitors of Apoptosis Proteins (IAPs), specifically targeting XIAP, cIAP1, and cIAP2. By mimicking the N-terminal IAP-binding motif of Smac, JP1201 binds to the BIR (Baculovirus IAP Repeat) domains of these proteins, thereby relieving the IAP-mediated inhibition of caspases and promoting apoptosis in cancer cells. Preclinical research has demonstrated that JP1201 can augment the efficacy of conventional chemotherapeutic agents, such as gemcitabine, in experimental models of pancreatic ductal adenocarcinoma (PDAC) and other solid tumors by overcoming IAP-mediated chemoresistance.
