JPCAR019 is an autologous chimeric antigen receptor T-cell (CAR-T) therapy being developed by Japan Tissue Engineering (J-TEC) for the treatment of acute lymphoblastic leukemia (ALL). The therapy consists of patient-derived T cells that are genetically engineered to express a chimeric antigen receptor targeting the CD19 antigen, which is highly expressed on B-cell malignancies. A distinguishing feature of JPCAR019 is its use of a non-viral, enzyme-based vector system (likely the piggyBac transposon system) for gene delivery, rather than the conventional lentiviral or retroviral vectors. This non-viral approach is designed to simplify the manufacturing process and reduce production costs. The program is currently in an investigator-initiated Phase 1/2 clinical trial in Japan, with potential future applications in other cancers and non-oncology indications.
