JPE 1375 is a peptidomimetic small molecule antagonist of the complement C5a receptors C5aR1 and C5aR2. Developed by Jerini AG, the compound is a hexapeptide derivative with the structure Hoo-Phe-Orn-Pro-hle-Pff-Phe-NH2. It functions by blocking the binding of C5a, a potent anaphylatoxin, to its receptors, thereby inhibiting the activation and mobilization of inflammatory cells such as polymorphonuclear leukocytes. Preclinical studies have demonstrated its ability to reduce TNF levels and show efficacy in models of peritonitis and dry age-related macular degeneration (AMD). However, its development has been characterized by a short in vivo duration of action of less than two hours.
