JPH203 (also known as Nanvuranlat or KYT-0353) is a first-in-class, selective small molecule inhibitor of L-type amino acid transporter 1 (LAT1). LAT1 is highly expressed on the cell membrane of many cancer cells and plays a critical role in the uptake of essential amino acids required for rapid tumor growth and proliferation. By competitively inhibiting LAT1, JPH203 disrupts amino acid transport into cancer cells, thereby suppressing their growth and survival. Preclinical studies have shown that JPH203 effectively inhibits leucine uptake and suppresses tumor cell proliferation. Clinically, it has demonstrated promising activity in patients with advanced biliary tract cancer refractory to standard therapies, with ongoing phase 2 trials. The drug was developed by J-Pharma and has received FDA orphan drug designation for biliary tract cancer[1][3][6][8].
