Drug intelligence / Profile preview
JS-K IP089 (also known as JS-K) is a first-in-class, small molecule nitric oxide (NO) donor belonging to the arylated diazeniumdiolate class. Developed by JSK Therapeutics in collaboration with the National Cancer Institute (NCI) and the University of Utah, JS-K is designed as a prodrug that is activated by glutathione S-transferases (GST)—enzymes frequently overexpressed in cancer cells—to release nitric oxide in a sustained and controlled manner. This selective release of NO impairs the redox state of malignant cells by depleting intracellular glutathione, inducing reactive oxygen species (ROS) accumulation, and triggering apoptosis and cell cycle arrest. JS-K has demonstrated potent, broad-spectrum antineoplastic and anti-angiogenic activity in preclinical models of various cancers, including prostate cancer, acute myeloid leukemia (AML), and multiple myeloma (MM).
