Drug intelligence / Profile preview
JS-K IPS8 is a preclinical-stage, first-in-class small molecule nitric oxide (NO) donor belonging to the O2-arylated diazeniumdiolate class, being developed by JSK Therapeutics for the treatment of various cancers, including ovarian cancer, acute myeloid leukemia (AML), and multiple myeloma. Designed as a prodrug, JS-K IPS8 is selectively activated by glutathione S-transferases (GST) in the presence of intracellular glutathione (GSH) to release cytotoxic levels of nitric oxide directly within malignant cells. This targeted release depletes intracellular glutathione, induces reactive oxygen species (ROS) accumulation, and triggers apoptosis and autophagy-dependent cell death while sparing normal tissues from systemic NO-mediated hypotension.
