Overview

JS007 is a novel humanized monoclonal antibody that targets cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4). By binding to CTLA-4, JS007 blocks its inhibitory signaling on T cells, thereby enhancing T cell activation and promoting antitumor immune responses. Preclinical studies demonstrated that JS007 has superior binding affinity and T-cell activating efficiency compared to ipilimumab. In vivo models showed increased tumor infiltration by effector T cells and depletion of regulatory T cells in the tumor microenvironment. A first-in-human phase 1a clinical trial in patients with advanced solid tumors indicated good tolerability, no dose-limiting toxicities up to 10 mg/kg IV every three weeks, a disease control rate of 50%, and a median overall survival of 14.7 months. The most common treatment-related adverse events were mild to moderate increases in liver enzymes; no severe immune-related adverse events were observed[1][5][2].
