Overview

JS011 is an investigational, orally bioavailable small molecule tyrosine kinase inhibitor (TKI) that selectively targets the Epidermal Growth Factor Receptor (EGFR) harboring exon 20 insertion mutations. Developed by Junshi Biosciences, JS011 is designed to address the significant unmet medical need in patients with non-small cell lung cancer (NSCLC) and other solid tumors whose genetic profiles include these specific mutations, which are typically resistant to first-, second-, and third-generation EGFR TKIs. By binding to the ATP-binding pocket of the mutated EGFR kinase domain, JS011 inhibits the activation of downstream signaling cascades, such as the PI3K/Akt/mTOR and RAS/RAF/MEK/ERK pathways, thereby suppressing tumor cell proliferation and survival. JS011 is currently undergoing Phase 1 clinical evaluation to determine its safety, tolerability, and preliminary efficacy in patients with advanced malignant solid tumors.
