JS015 is a recombinant humanized anti-DKK1 monoclonal antibody developed by Shanghai Junshi Biosciences. It is designed to target and inhibit Dickkopf-related protein 1 (DKK1), a key negative regulator of the Wnt signaling pathway. By binding to DKK1 in the tumor microenvironment, JS015 blocks its interaction with LRP5/6 receptors, thereby activating Wnt signaling. This activation can promote anti-tumor immune responses by stimulating natural killer (NK) cells and T lymphocytes while inhibiting myeloid-derived suppressor cells (MDSCs). The drug is being investigated primarily for advanced malignant solid tumors, including colorectal cancer, gastroesophageal junction adenocarcinoma, esophageal squamous cell carcinoma, pancreatic ductal adenocarcinoma, rectal cancer, and stomach adenocarcinoma[1][2][3].
