Overview

JS104 is a monoclonal antibody developed by Junshi Biosciences as a biosimilar to cetuximab (Erbitux). It targets the epidermal growth factor receptor (EGFR), a transmembrane glycoprotein that is overexpressed in several types of solid tumors, including colorectal and head and neck cancers. By binding to the extracellular domain of EGFR, JS104 prevents the binding of natural ligands such as epidermal growth factor (EGF) and transforming growth factor-alpha (TGF-α), thereby inhibiting the activation of downstream signaling cascades (e.g., RAS-MAPK and PI3K-Akt) that drive tumor cell proliferation, survival, and metastasis. Additionally, JS104 can trigger antibody-dependent cellular cytotoxicity (ADCC), leading to the destruction of EGFR-expressing tumor cells by the immune system. JS104 is currently in Phase 3 clinical trials for the treatment of metastatic colorectal cancer.
