JS105 + fluzoparib is a combination therapy consisting of JS105, an orally bioavailable, selective inhibitor of the PI3Kα isoform, and fluzoparib, a potent PARP1/2 inhibitor. Developed through a collaboration between Junshi Biosciences and Jiangsu Hengrui Pharmaceuticals, this combination targets the synergistic relationship between the PI3K/AKT pathway and DNA damage repair mechanisms. PI3Kα inhibition has been shown to downregulate the expression of BRCA1/2 and other homologous recombination (HR) genes, thereby inducing a state of functional HR deficiency (BRCAness) that sensitizes tumor cells to PARP inhibition. This combination is primarily being investigated for the treatment of advanced solid tumors, particularly those harboring PIK3CA mutations, including breast and gynecological cancers.
