JS111 is an investigational small molecule inhibitor targeting the epidermal growth factor receptor (EGFR), specifically designed to inhibit uncommon EGFR mutations in non-small cell lung cancer (NSCLC). These mutations include EGFR exon 20 insertions, T790M point mutation, complex mutations, and other point or sequence repeat mutations between exons 18 and 21 such as G719X. JS111 has demonstrated preclinical activity against both common and uncommon EGFR mutations and aims to address the limited efficacy of existing EGFR tyrosine kinase inhibitors (TKIs) for patients with these rare alterations. The drug is being developed as an oral therapy for advanced NSCLC harboring EGFR mutations[2][5][6].
