Overview

JS2-DNA is an experimental DNA plasmid vaccine candidate developed for the prevention and treatment of HIV-1 infection. The vaccine consists of a plasmid vector encoding structural and enzymatic proteins—specifically Gag, Pol, and Env—derived from the HIV-1 subtype B JS2 isolate, which is a primary isolate from an Australian patient. The mechanism of action involves the uptake of the DNA plasmid by host cells following intramuscular injection, leading to the endogenous expression of these viral antigens. These proteins are then processed and presented to the immune system, primarily to induce virus-specific cellular immune responses, including CD4+ and CD8+ T-cell activation, as well as humoral responses. JS2-DNA has been evaluated in early-phase clinical trials, frequently as a priming component in heterologous prime-boost regimens, often paired with a recombinant viral vector boost such as Modified Vaccinia Ankara (MVA).
