JS203 + lenalidomide is a combination therapy being developed by Shanghai Junshi Bioscience for the treatment of various B-cell non-Hodgkin lymphomas, including diffuse large B-cell lymphoma (DLBCL), follicular lymphoma (FL), mantle cell lymphoma (MCL), and marginal zone lymphoma (MZL). JS203 is a recombinant humanized bispecific antibody that targets CD20 on malignant B-cells and CD3 on T-cells, facilitating T-cell-mediated cytotoxicity against the tumor cells. Lenalidomide is an oral immunomodulatory drug (IMiD) that binds to the Cereblon (CRBN) E3 ubiquitin ligase complex, leading to the degradation of transcription factors Ikaros (IKZF1) and Aiolos (IKZF3). This action results in direct anti-tumor effects and enhances the activity of immune effector cells, potentially providing a synergistic therapeutic effect when combined with the T-cell engaging mechanism of JS203.
