Overview

JS301 is a bispecific antibody-drug conjugate (ADC) developed by TopAlliance Biosciences (a subsidiary of Junshi Biosciences) that simultaneously targets Human Epidermal Growth Factor Receptor 2 (HER2) and Carbonic Anhydrase IX (CAIX). This dual-targeting approach is intended to increase the precision of cytotoxic payload delivery to tumor cells, particularly in cancers where these two antigens are co-expressed or where CAIX is upregulated due to tumor hypoxia. By binding to HER2 and CAIX, JS301 is internalized into the cancer cell, where the linker is cleaved to release a potent cytotoxic agent, leading to cell cycle arrest and apoptosis. The drug is currently in early-stage clinical development for the treatment of advanced solid tumors.
