JSI-1187

JSI-1187 is a selective, orally administered small molecule inhibitor targeting ERK1 and ERK2 (extracellular-signal-regulated kinases 1 and 2), key components of the MAPK signaling pathway. It is being developed primarily for the treatment of advanced solid tumors harboring mutations that activate the MAPK pathway, including BRAF and RAS mutations. Preclinical studies have shown high potency against various tumor types with these mutations. Clinical development includes both monotherapy and combination therapy (notably with dabrafenib) in patients with locally advanced or metastatic solid tumors such as melanoma and non-small cell lung cancer (NSCLC). The drug is currently in phase 1 clinical trials in both the United States and China[1][4][5][8].
