JSKN016 + carboplatin is a **combination therapy** consisting of JSKN016, a bispecific antibody-drug conjugate (ADC), and carboplatin, an alkylating agent. JSKN016 targets both HER3 (Human epidermal growth factor receptor 3) and TROP2 (Trophoblast cell surface antigen 2), which are overexpressed in various solid tumors. JSKN016 binds these antigens on tumor cells and undergoes internalization and lysosomal processing to release a topoisomerase I inhibitor (TOPIi) payload, leading to apoptosis. JSKN016 further exerts **bystander effect** by enabling its payload to enter antigen-negative cells. Carboplatin is a platinum-based chemotherapeutic agent that creates DNA cross-links and induces cell death through cell-cycle nonspecific mechanisms. The combination is under active clinical investigation for the treatment of advanced solid tumors, including non-small cell lung cancer (NSCLC) and HER2-negative breast cancer. JSKN016 is developed by Alphamab Oncology, and studies of the combination are ongoing in China[2][3][4][5][7][9][10][8].
