JSKN016 + carboplatin + ivonescimab is a triple combination therapy currently being investigated in clinical trials primarily for advanced malignant solid tumors, HER2-negative breast cancer, and non-small cell lung cancer. JSKN016 is a bispecific antibody-drug conjugate (ADC) targeting HER3 and TROP2, which delivers a cytotoxic topoisomerase I inhibitor (TOPIi) upon binding, causing tumor cell apoptosis and exhibiting a bystander effect on antigen-negative cells[1][2][4]. Carboplatin is a platinum-based chemotherapeutic agent that induces DNA crosslinking, leading to cell death. Ivonescimab is a bispecific monoclonal antibody targeting PD-1 and VEGF, functioning as a checkpoint inhibitor with anti-angiogenic properties. This combination is designed to simultaneously target tumor cell receptors, induce DNA damage, and suppress immune checkpoint and angiogenesis pathways for enhanced antitumor effect.
