JSKN016 + docetaxel is an investigational combination therapy composed of JSKN016, a bispecific antibody-drug conjugate (ADC) targeting HER3 (Human epidermal growth factor receptor 3) and TROP2 (Trophoblast cell surface antigen 2), and docetaxel, a well-established chemotherapy agent. JSKN016 is developed using Alphamab Oncology’s proprietary Glycan-specific conjugation platform. Upon binding to TROP2 and/or HER3 on tumor cells, JSKN016 is internalized via endocytosis, releasing a cytotoxic topoisomerase I inhibitor (TOPIi), which triggers apoptosis in TROP2/HER3-positive tumor cells and presents a bystander effect by affecting adjacent antigen-negative tumor cells. Docetaxel is a cytotoxic taxane chemotherapy that inhibits cell division by stabilizing microtubules. The combination is being evaluated in clinical studies for efficacy and safety in advanced solid tumors, including non-small cell lung cancer (NSCLC) and HER2-negative breast cancer[1][2][3][4][5].
