JSKN016 + pembrolizumab is a combination investigational anticancer regimen consisting of JSKN016, a bispecific antibody-drug conjugate (ADC) targeting both HER3 (human epidermal growth factor receptor 3) and TROP2 (trophoblast cell surface antigen 2), and pembrolizumab, a programmed cell death protein 1 (PD-1) immune checkpoint inhibitor. JSKN016 binds HER3 and/or TROP2 on tumor cells, is internalized via endocytosis, and releases a cytotoxic topoisomerase I inhibitor payload, inducing apoptosis and also exerting a bystander effect on antigen-negative cells[1][2][3][4][6][8]. Pembrolizumab blocks interaction of PD-1 with its ligands, restoring immune response against tumor cells[9]. The combination is currently being studied for synergistic immunotherapeutic and cytotoxic activity primarily in advanced HER2-negative breast cancer and non-small cell lung cancer (NSCLC)[3][4][6].
