JSKN016SC is a subcutaneous formulation of JSKN016, a bispecific antibody-drug conjugate (ADC) developed by Alphamab Oncology (Jiangsu Alphamab Biopharmaceuticals). It simultaneously targets Human Epidermal Growth Factor Receptor 2 (HER2) and Human Epidermal Growth Factor Receptor 3 (HER3). The drug utilizes Alphamab's proprietary glycan-fixation site-specific conjugation platform to attach a cytotoxic payload, typically a topoisomerase I inhibitor, to the bispecific antibody. This dual-targeting approach is designed to enhance tumor specificity and overcome resistance mechanisms associated with single-target HER2 or HER3 therapies. The subcutaneous administration route is being evaluated for its potential to improve patient convenience and provide a distinct pharmacokinetic profile compared to traditional intravenous ADC delivery. It is currently in early-phase clinical development for advanced solid tumors, including non-small cell lung cancer (NSCLC) with EGFR mutations and HER2-negative breast cancer.
