JSKN021 is a first-in-class dual-payload bispecific antibody-drug conjugate (ADC) developed by Alphamab Oncology. It targets both EGFR and HER3, utilizing a bispecific antibody backbone engineered for finely tuned binding avidity to address tumor heterogeneity and minimize off-tumor toxicity. The drug is conjugated with two distinct payloads: a novel topoisomerase I inhibitor (T01) with a drug-to-antibody ratio (DAR) of 4, and monomethyl auristatin E (MMAE) with a DAR of 2. This dual-payload strategy is designed to overcome resistance and non-response associated with single-payload ADCs. JSKN021 employs glycan-specific conjugation and the Alphatecan® hydrophilic cleavable linker system. It is currently in preclinical development for various oncology indications.
