Overview

JSKN029 is a preclinical-stage antibody-drug conjugate (ADC) being developed by AlphaMab Oncology. It is designed to target Nectin-4 (Poliovirus receptor-related protein 4), a cell surface adhesion molecule that is significantly overexpressed in various solid tumors, including urothelial, breast, and lung cancers, while maintaining limited expression in healthy tissues. JSKN029 utilizes AlphaMab's proprietary glycan-site-specific conjugation platform, which typically involves the stable attachment of a potent cytotoxic payload—often a topoisomerase I inhibitor—to a monoclonal antibody. This targeted approach is intended to deliver the cytotoxic agent directly to Nectin-4-expressing malignant cells, thereby inducing apoptosis while minimizing systemic side effects and expanding the therapeutic window compared to traditional chemotherapy.
