Overview

JSM6427 is a synthetic peptide and small molecule integrin alpha5beta1 antagonist developed primarily for ophthalmologic indications. It acts by selectively inhibiting the interaction between integrin α5β1 (the fibronectin receptor) and its ligand, thereby blocking multiple angiogenic growth factor pathways involved in pathological neovascularization, inflammation, and fibrosis. Preclinical studies demonstrated that intravitreal administration of JSM6427 inhibits choroidal neovascularization (CNV), reduces retinal cell proliferation, and suppresses inflammatory responses in animal models. Clinical development focused on age-related macular degeneration (AMD) and proliferative vitreoretinopathy; however, the program was discontinued after Phase 1 trials due to strategic reasons rather than safety concerns. The drug was originally developed by Jerini Ophthalmic (a subsidiary of Jerini AG), later acquired by Shire.
