Overview

JT118 is a recombinant protein vaccine candidate developed for the prevention of monkeypox virus infection[1][3][5][7][11][13][15][16]. It consists of a "two-in-one" tandem fusion protein combining two monkeypox virus antigens: **A35** (an extracellular enveloped virus antigen) and **M1** (an intracellular mature virus antigen)[1][3][7]. This structure is designed to retain the natural epitope structure of both antigens and increase the molecular weight, thereby enhancing immunogenicity. Preclinical data show that JT118 induces high-titer binding and neutralizing antibodies against both vaccinia and monkeypox viruses, as well as robust cellular immune responses in animal models[1][3][4][10]. These immune responses translate into significant protection against viral challenge with a favorable safety profile[1][3][10]. The vaccine is being developed by Shanghai JunTop Biosciences, a subsidiary of Shanghai Junshi Biosciences, in collaboration with Peking University, the Institute of Microbiology (Chinese Academy of Sciences), Shanxi Academy of Advanced Research and Innovation, and Beihang University[3]. As of June 2025, JT118 has received regulatory acceptance for clinical trial registration in China, with IND approval granted for Phase 1 studies[1][3][11][15][16].
