Overview

**JTCR016** is an adoptive T lymphocyte immunotherapy utilizing augmented T-cell receptor (TCR) technology to target Wilms' tumor 1 (WT1), an intracellular protein overexpressed in various hematologic malignancies and solid tumors. Developed originally by Juno Therapeutics and now under Bristol Myers Squibb following acquisitions, it involves engineering patient T cells with a high-affinity TCR specific to HLA-A*02:01-restricted WT1 peptides, enabling recognition and immunologic cytotoxicity against WT1-expressing cancer cells. It has been evaluated in Phase I/II trials for cancers including acute myeloid leukemia (AML), chronic myeloid leukemia (CML), myelodysplastic syndromes (MDS), non-small cell lung cancer (NSCLC), mesothelioma, and precursor cell lymphoblastic leukemia-lymphoma, showing preliminary efficacy such as partial responses and stable disease with a tolerable safety profile lacking severe cytokine release syndrome or neurotoxicity.[1][4][5][6][7][10][14]
