JTE-051

JTE-051 is an orally available small molecule inhibitor of interleukin-2-inducible T cell kinase (ITK), a member of the Tec family of non-receptor tyrosine kinases that plays a critical role in T cell activation and immune response. Developed by Japan Tobacco, with Akros Pharma also involved, JTE-051 has been investigated as an antiallergic, antipsoriatic, and antirheumatic agent. Its primary mechanism is the inhibition of ITK signaling pathways, thereby suppressing T cell activation implicated in autoimmune and inflammatory diseases such as rheumatoid arthritis, plaque psoriasis, and hypersensitivity disorders. Clinical trials have reached Phase II for these indications.
