Overview
JTE-451 is a small molecule antagonist of the orphan nuclear receptor RORγ (nuclear receptor subfamily 1 group F member 3). It was developed as an antiallergic and antipsoriatic agent, primarily targeting the RORγ pathway. The drug was investigated for the treatment of plaque psoriasis, with both oral and topical formulations evaluated in clinical trials. Its mechanism of action involves antagonism of RORγ, which plays a key role in immune regulation and inflammation. The originator is Orphagen Pharmaceuticals, with development led by Japan Tobacco. Clinical development has been discontinued for plaque psoriasis[1][2][3][8].
