JTK-003

JTK-003 is an orally active small molecule glucokinase activator (GKA) that was developed by Japan Tobacco for the treatment of type 2 diabetes mellitus. Glucokinase (GCK) serves as a critical glucose sensor in the body, primarily expressed in pancreatic beta cells and hepatocytes, where it regulates insulin secretion and hepatic glucose metabolism. By allosterically activating GCK, JTK-003 enhances glucose-stimulated insulin secretion from the pancreas and promotes glucose phosphorylation and glycogen synthesis in the liver, thereby lowering blood glucose levels. Clinical development of JTK-003 reached Phase 2 trials, where its efficacy and safety were evaluated in patients with type 2 diabetes; however, the program was subsequently discontinued by the developer.
