JTV-261 is an orally active small molecule inhibitor of the sodium-glucose cotransporter 1 (SGLT1), developed by Japan Tobacco Inc. SGLT1 is the primary transporter for glucose and galactose absorption in the small intestine and is also expressed in the S3 segment of the renal proximal tubule. By selectively inhibiting SGLT1, JTV-261 is designed to delay the absorption of dietary glucose, thereby reducing postprandial glucose excursions and potentially stimulating the release of glucagon-like peptide-1 (GLP-1). This mechanism distinguishes it from SGLT2 inhibitors, which primarily act on renal glucose reabsorption. JTV-261 is currently being evaluated in Phase 1 clinical trials for safety, tolerability, and pharmacokinetics in healthy volunteers, with potential applications in metabolic disorders such as type 2 diabetes.
