JTX-8064 is a humanized IgG4 monoclonal antibody that specifically targets and binds to Leukocyte Immunoglobulin Like Receptor B2 (LILRB2, also known as ILT4), an immune checkpoint molecule primarily expressed on myeloid cells such as monocytes and macrophages. By blocking the interaction between LILRB2 and its ligands—including classical and non-classical major histocompatibility complex class I (MHC I) molecules—JTX-8064 inhibits LILRB2-mediated signaling. This blockade reprograms immunosuppressive (M2) macrophages toward a pro-inflammatory (M1) phenotype, thereby enhancing antigen presentation, T cell activation, and overall anti-tumor immunity. JTX-8064 is being developed for the treatment of advanced solid tumors both as monotherapy and in combination with PD-1 inhibitors such as pimivalimab. The drug was originally developed by Jounce Therapeutics; development has since continued under Concentra Biosciences[1][3][6][7][8].
