Overview

JV-394 is an autologous, second-generation chimeric antigen receptor (CAR) T-cell therapy targeting CD94 (Killer cell lectin-like receptor subfamily D member 1). Developed by M.D. Anderson Cancer Center, the therapy is designed for the treatment of relapsed or refractory CD94-positive T/NK cell neoplasms, including extranodal NK/T-cell lymphoma, hepatosplenic T-cell lymphoma, and aggressive NK cell leukemia. The CAR construct features a single-chain variable fragment (scFv) derived from a monoclonal antibody and a 4-1BB (CD137) co-stimulatory domain, which promotes T-cell expansion and persistence. CD94 is a C-type lectin receptor highly expressed in various aggressive T and NK cell malignancies, making it a specific target for directed immunotherapy.
