JV101 is an investigational adeno-associated virus (AAV) gene therapy developed by Juvensis Therapeutics for the treatment of heart failure, specifically dilated cardiomyopathy (DCM) and heart failure with preserved ejection fraction (HFpEF). The therapy utilizes an AAV9 vector to deliver a modified version of the telomerase reverse transcriptase (TERT) gene, driven by a cardiac troponin T (cTnT) promoter to ensure heart-specific expression. By targeting the telomerase pathway, JV101 aims to restore telomere length in cardiomyocytes and improve mitochondrial biogenesis, thereby addressing cellular aging and dysfunction associated with heart failure. It is currently in early-phase clinical development, with an exploratory Phase 1 trial evaluating its safety and feasibility in patients with heart failure due to dilated cardiomyopathy.
