JVRS-100 is a cationic lipid-DNA complex (CLDC) developed as an adjuvant to enhance the immunogenicity and efficacy of vaccines, particularly for influenza and other infectious diseases. The formulation consists of DOTIM/cholesterol liposomes combined with plasmid DNA containing immunostimulatory CpG and non-CpG motifs. Its mechanism of action involves self-assembly with disease-specific antigens, leading to robust antibody and cell-mediated immune responses—especially induction of CD4+ and CD8+ T lymphocytes. Preclinical studies have shown that adding JVRS-100 to inactivated H5N1 influenza vaccines significantly increases hemagglutination-inhibition titers, neutralizing antibodies, cross-protection against multiple viral clades, and protection from lethal challenge in animal models. Clinical development has focused on its use as an adjuvant for seasonal influenza vaccines in elderly populations as well as potential applications in cancer immunotherapy[1][2][5][6][8].
