JW 1 (also referred to as JW-1) is an experimental small molecule inhibitor of Janus kinase 3 (JAK3) that has been evaluated in preclinical models for the treatment of inflammatory skin diseases, primarily psoriasis. The compound targets the JAK-STAT signaling pathway by selectively inhibiting JAK3, an enzyme essential for the signaling of cytokines that utilize the common gamma chain (CD132), including IL-2, IL-4, IL-7, IL-9, IL-15, and IL-21. By blocking these pathways, JW 1 suppresses the activation and proliferation of T-cells and reduces the production of pro-inflammatory mediators such as IL-17 and interferon-gamma. Preclinical evaluations, including studies conducted by the contract research organization Derphartox using a human skin transplant model of psoriasis, have demonstrated that topical application of JW 1 can significantly reduce epidermal hyperplasia and inflammatory cell infiltration.
