JW-1-283 is a small molecule inhibitor designed to target the interaction between the MDM2 E3 ubiquitin-protein ligase and the p53 tumor suppressor protein. Developed by researchers at the University of Tennessee Health Science Center, it aims to reactivate the p53 pathway, which is frequently inactivated in melanoma despite being wildtype in approximately 90% of cases. Mechanistically, JW-1-283 disrupts the p53-MDM2 complex, preventing the proteasomal degradation of p53 and leading to its stabilization. This stabilization is accompanied by p38-mediated phosphorylation of p53, resulting in S-phase cell cycle arrest and apoptosis in cancer cells. Preclinical studies in A375 melanoma models have demonstrated significant inhibition of tumor growth, reduced stemness, and impaired angiogenesis.
