Overview

JW-1521 is a small molecule tubulin polymerization inhibitor currently under development by San Rocco Therapeutics (formerly Errant Gene Therapeutics) for the treatment of refractory prostate cancer. Originally discovered by JW Pharmaceutical, the compound acts by binding to the colchicine binding site on tubulin, which disrupts microtubule assembly and leads to cell cycle arrest at the G2/M phase and subsequent apoptosis. JW-1521 is specifically designed to address the clinical need in patients with castration-resistant prostate cancer (CRPC) who have developed resistance to taxane-based chemotherapies, such as docetaxel. Unlike taxanes, which bind to the taxane site and stabilize microtubules, JW-1521's mechanism of destabilization and its ability to bypass P-glycoprotein-mediated efflux make it a candidate for overcoming multi-drug resistance. It is currently being evaluated in Phase 1 clinical trials.
