Overview

JW-3 is an experimental small molecule inhibitor of **p38 mitogen-activated protein kinase (p38 MAPK)**, belonging to the pyridinyl imidazole chemical class. It was developed and evaluated in the early 2000s as part of a series of test compounds (JW-1, JW-2, and JW-3) for the treatment of inflammatory skin conditions, most notably **psoriasis**. Preclinical studies, including those conducted by the dermatology-focused CRO **Derphartox** in a human skin transplant model, demonstrated that JW-3 effectively reduces the expression of pro-inflammatory cytokines (such as TNF-alpha and IL-1 beta) and normalizes keratinocyte proliferation. The compound is structurally related to the prototypical p38 inhibitor SB 203580 and was primarily used as a research tool to investigate the therapeutic potential of the MAPK pathway in psoriatic lesions.
