Overview

JW-65 is a potent, selective, and brain-penetrant small-molecule inhibitor of transient receptor potential canonical 3 (TRPC3) channels. Developed by SEAK Therapeutics in collaboration with researchers at the University of Tennessee Health Science Center (UTHSC), JW-65 was optimized from the prototypic TRPC3 inhibitor pyrazole 3 (Pyr3) to improve metabolic stability and reduce toxicity. It is being investigated for the treatment of epilepsy (including inherited generalized tonic-clonic epilepsy and chemoconvulsant-induced seizures) and Alzheimer's disease, where it has demonstrated neuroprotective effects by mitigating calcium overload and reducing beta-amyloid-induced synaptic dysfunction.
