JW02 is an allogeneic (universal) chimeric antigen receptor (CAR) T-cell therapy targeting C-type lectin-like molecule-1 (CLL1, also known as CLEC12A). Developed by Juventas Cell Therapy, it is currently in Phase 1 clinical development for the treatment of acute myeloid leukemia (AML). As a "universal" or "off-the-shelf" product, JW02 utilizes T cells from healthy donors that are genetically modified to express a CAR specific for CLL1 while undergoing gene editing to minimize the risk of graft-versus-host disease (GvHD) and immune rejection. CLL1 is a promising target in AML as it is highly expressed on leukemic blasts and leukemic stem cells but has restricted expression on normal hematopoietic stem cells, potentially allowing for the eradication of the disease while sparing the bone marrow's regenerative capacity.
