Overview

JWATM204 is an autologous CAR-T cell therapy targeting glypican-3 (GPC-3), a protein overexpressed in hepatocellular carcinoma (HCC) and other solid tumors. Developed using the ARTEMIS T-cell platform from Eureka and in partnership with Lyell, JWATM204 utilizes high-affinity, GPC-3-specific monoclonal antibody engineering and enhanced co-stimulatory signaling to promote T cell activation and antitumor effects. It is designed for patients with advanced HCC and is currently in early-phase clinical evaluation. Preclinical studies showed promising anti-tumor activity, leading to ongoing first-in-human studies in adult patients with advanced, GPC-3-expressing HCC. The main mechanism is T-cell mediated cytotoxicity via CAR recognition of GPC-3 on tumor cells, potentially offering a novel therapeutic approach for patients refractory to standard treatments[1][3][5][7][8].
