Overview

JWK013 is an investigational gene therapy developed by West China Hospital, Sichuan University, for the treatment of treatment-resistant depressive disorder (TRD). It utilizes an adeno-associated virus serotype 9 (AAV9) vector to deliver the *NRF2* (Nuclear factor erythroid 2-related factor 2) gene via intranasal administration. NRF2 is a transcription factor that serves as a master regulator of the antioxidant response, inducing the expression of numerous genes that protect against oxidative stress and neuroinflammation. By overexpressing NRF2 in the brain, JWK013 aims to mitigate the inflammatory and oxidative damage implicated in the pathophysiology of depression. The therapy is currently being evaluated in a Phase 1 dose-escalation clinical trial in China to assess its safety, tolerability, and preliminary antidepressant effects in adults with TRD.
