JY03 is a recombinant humanized monoclonal antibody developed by Beijing Dongfang Baitai Biotechnology as a biosimilar to pertuzumab (Perjeta). It specifically targets the extracellular dimerization domain (subdomain II) of the Human Epidermal Growth Factor Receptor 2 (HER2). By binding to this site, JY03 prevents HER2 from forming heterodimers with other members of the HER family, such as HER1 (EGFR), HER3, and HER4. This inhibition disrupts downstream signaling through the mitogen-activated protein kinase (MAPK) and phosphoinositide 3-kinase (PI3K) pathways, which are critical for the growth and survival of HER2-overexpressing cancer cells. JY03 is primarily being evaluated for the treatment of HER2-positive solid tumors, including breast cancer, often in combination with other anti-HER2 therapies like trastuzumab.
