JYP0035 + fulvestrant is an investigational combination therapy being developed by Chengdu JOYO Pharma for the treatment of advanced breast cancer. The regimen consists of JYP0035, a small molecule inhibitor of the alpha isoform of phosphoinositide 3-kinase (PI3Kα), and fulvestrant, a selective estrogen receptor degrader (SERD). This combination is specifically targeted at patients with hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) advanced breast cancer that harbors PIK3CA mutations and has progressed on prior endocrine therapy. By simultaneously inhibiting the PI3K/AKT/mTOR pathway and degrading the estrogen receptor, the therapy aims to counteract crosstalk between these pathways that often leads to treatment resistance. It is currently being evaluated in Phase III clinical trials.
