JZY-2233 is a first-in-class degrader-antibody conjugate (DAC) designed for the treatment of advanced prostate cancer. It consists of a potent small-molecule degrader targeting the transcriptional coactivators CBP (CREB-binding protein) and p300, conjugated to a monoclonal antibody specific for prostate-specific membrane antigen (PSMA). By leveraging the tumor-specific expression of PSMA, JZY-2233 delivers the CBP/p300 degrader directly to prostate cancer cells, where it induces the proteasomal degradation of these essential androgen receptor coactivators. This targeted degradation suppresses downstream oncogenic drivers, including c-Myc and PSA, leading to robust growth inhibition. Preclinical studies in xenograft models have demonstrated that JZY-2233 provides prolonged tumor suppression with significantly reduced systemic toxicity compared to unconjugated CBP/p300 degraders.
