Overview

K-232 is a Rho-kinase (ROCK) inhibitor developed by Kowa Company, Ltd. for the treatment of primary open-angle glaucoma, exfoliative glaucoma, pigmentary glaucoma, and ocular hypertension. It is a new formulation of ripasudil hydrochloride hydrate (originally developed as K-115) optimized for once-daily ophthalmic administration. By inhibiting ROCK1 and ROCK2, K-232 promotes the outflow of aqueous humor through the trabecular meshwork, thereby reducing intraocular pressure. As of 2024, it is in Phase 3 clinical development in Japan, where it is being evaluated for long-term safety and efficacy both as a monotherapy and in combination with other intraocular pressure-lowering agents.
