Overview

K-319 is a recombinant fusion protein comprising the Fc regions of human IgE and IgG, specifically designed for the treatment of allergic asthma and other IgE-mediated allergic diseases. Developed by Shanghai Kexin Biopharmaceutical, the molecule functions by simultaneously binding to the high-affinity IgE receptor (FcεRI) and the inhibitory IgG receptor (FcγRIIb) on the surface of mast cells and basophils. This co-aggregation triggers the phosphorylation of the immunoreceptor tyrosine-based inhibitory motif (ITIM) within the cytoplasmic tail of FcγRIIb, which recruits phosphatases such as SHIP to inhibit the signaling cascade initiated by FcεRI. Consequently, K-319 suppresses the degranulation of these cells and the subsequent release of histamine and other pro-inflammatory mediators, offering a targeted approach to dampening allergic responses.
