Overview

K-552 is a selective inhibitor of Ubiquitin-specific protease 47 (USP47). USP47 is known to stabilize the oncoprotein c-Myc by preventing its proteasomal degradation through deubiquitination, which promotes non-small cell lung cancer (NSCLC) cell proliferation. By inhibiting USP47, K-552 effectively destabilizes c-Myc, thereby inhibiting KRASG12C-mutated NSCLC cell proliferation. Furthermore, K-552 has been shown to enhance the efficacy of KRASG12C inhibitors, such as Sotorasib, both in vitro and in vivo, suggesting its potential as a combination therapy for KRASG12C-mutated NSCLC.
