K-554 (Kallyope)

K-554 is a first-in-class, once-weekly injectable small molecule therapeutic candidate developed by Kallyope for the treatment of obesity and metabolic diseases. It targets an undisclosed, novel non-incretin peptide receptor that is central to a neural circuit involved in the regulation of feeding behavior. Unlike current obesity treatments that focus on GLP-1 or amylin pathways, K-554 acts through a distinct mechanism associated with non-aversive satiety signals, potentially offering a differentiated tolerability profile. The program is currently in Phase 1 development, with first-in-human studies planned to initiate in mid-2026.
