Overview

K-833 is a novel, gut-restricted small molecule that acts as a full agonist of the G-protein coupled receptor 119 (GPR119). It is being developed as an oral treatment for obesity and type 2 diabetes mellitus in obese patients. The drug enhances the body's natural metabolic signals to stimulate the secretion of multiple appetite-suppressing satiety hormones, including GLP-1 (glucagon-like peptide 1), PYY (peptide YY), and CCK (cholecystokinin). This mechanism mimics hormone release patterns seen after bariatric surgery and aims to promote weight loss and improve glycemic control. Unlike GLP-1 receptor agonists currently on the market, K-833 targets nutrient receptors in the gut to trigger these effects. Clinical studies have shown that when used in combination with another agent (K-757), it can lead to modest but statistically significant weight loss with improved tolerability compared to some injectable therapies[1][3][4][6][8].
